On August 19, JPMorgan released a research report indicating that Moderna and Merck's personalized neoantigen therapy (INT) met the primary endpoint in adjuvant melanoma, with a significant improvement in relapse-free survival, and also met the key secondary endpoint of distant metastasis-free survival. However, this outcome was in line with expectations, and the market had already priced it into the stock before the announcement.
Following the news, Moderna's stock rose in pre-market trading. JPMorgan maintained its Underweight rating and $40 price target, implying a 36% downside from the current price of around $63. The core rationale is: the success in adjuvant melanoma has already been priced in, and Moderna's next phase of value depends on whether INT can replicate this success in broader indications.
Melanoma Results Align with 85% Success Expectation
JPMorgan had previously assigned an 85% probability of success to the INT Phase III clinical trial in adjuvant melanoma. This assessment was based on the strong efficacy signal shown in the Phase IIb data and the biological plausibility of the mechanism in tumor immunotherapy.
Relapse-free survival was the primary endpoint, and distant metastasis-free survival was the key secondary endpoint. JPMorgan cited a melanoma expert's view, stating that distant metastasis-free survival is an "extremely meaningful endpoint" and "where the value lies." The appearance of distant metastases often signals a significant worsening of prognosis, and delaying or preventing distant metastases has meaningful clinical substance for improving patients' quality of life.
The report concludes that this result itself is not an incremental driver for the stock price. Moderna's market capitalization approached $25 billion before the announcement, and JPMorgan believes this already incorporated expectations for INT's approval in adjuvant melanoma. After risk adjustment, this indication contributes only about 3% to JPMorgan's valuation.
INT's Cross-Indication Data is the Key Variable
Adjuvant melanoma treatment represents a relatively small indication within the field of tumor immunotherapy. The combination of INT and Merck's Keytruda demonstrates clear clinical value, but the market size is limited. For Moderna to support a $25 billion market capitalization, INT needs to be similarly effective in other indications.
JPMorgan's valuation model already includes approximately $15 per share in risk-adjusted valuation for INT's potential value in other indications. The report remains focused on data disclosures and subsequent trials for INT in non-melanoma indications, as these are the key factors influencing the valuation assessment.
Adjuvant melanoma is the most advanced indication in the INT pipeline, but it is far from the end. Clinical developments in other cancer types will determine the ceiling of INT as a platform technology. From a biological mechanism perspective, the logic of personalized neoantigen therapy is not limited to melanoma; it theoretically holds feasibility in other immunogenic tumor types such as lung cancer, head and neck cancer, and renal cell carcinoma. However, theoretical feasibility does not equal clinical validation; subsequent data is key.
Valuation Logic Behind the $40 Price Target
JPMorgan's $40 price target is based on a DCF valuation model using a 10.5% weighted average cost of capital and a 0% terminal growth rate. This valuation covers Moderna's currently commercialized products and its risk-adjusted pipeline value.
Upside risks include INT data in non-melanoma indications exceeding expectations, faster product approval timelines, and improved vaccine demand driven by policy changes. Downside risks are typical pharmaceutical risks faced by Moderna in mRNA platform competition: clinical failures, regulatory delays, and commercial uptake falling short of expectations.
The pre-market rise reflects more of a short-covering and short-term trading sentiment rather than a structural change in fundamentals. INT's performance in other indications determines the valuation ceiling. If subsequent data can prove this platform technology has clinical activity in more tumor types, Moderna's valuation will see a significant upward revision. If data falls short of expectations, the portion of cross-indication value already priced into the current stock price will face contraction.
JPMorgan maintains its bearish stance. The clinical value of INT in melanoma is clear but has been fully priced in. Moderna needs to prove that INT is similarly effective in more cancer types.
